[pharmacology and toxicology] Artemether in this product is a derivative of artemisinin, which has a strong and rapid killing effect on the internal stage of plasmodium, and can quickly control the clinical onset and symptoms.The mechanism of action of artemisinin is still not clear. It mainly interferes with the mitochondrial function of the plasmodium membrane. Artemisinin changes the membrane structure of plasmodium by affecting the ultrastructure of plasmodium red.Due to the effect on the food bubble membrane, the nutrient intake of the plasmodium is blocked, and when the plasmodium loses a large amount of cytoplasm and nutrients, it is not replenished, so it dies quickly. The mechanism of action is through its internal peroxide (dioxygen) bridge, mediated by free iron from hemoglobin decomposition, which produces unstable organic free radicals and/or other electricphilic mediators, which then form a covalent admixture with the protein of the plasmodium, killing the plasmodium.Artemether has six times more antimalarial activity than artemisinin. The LD50 of artemether orally in mice was 895 ~ 977mg/kg, and that of artemether oil intramuscular in rats was 597mg/kg.Subacute toxicity test showed that after intramuscular injection of artemether oil at 18mg/kg×14 days, rats had no adverse reactions except a decrease in the number of reticulocytes. This drug has certain embryonic toxicity, which is manifested as embryonic absorption. Benzofluorene alcohol can kill plasmodium red interphase schizozoite completely, and the cure rate is about 95%, but it is not effective for erythrocyte prophase and gametophyte.
