ABclonal Second Strand Synthesis Module (Stranded) (RK20343) generates a double-stranded cDNA product using the product from ABclonal First strand cDNA synthesis as a template for second strand cDNA synthesis. This double-stranded cDNA product can be used directly for DNA construction.
The Second Strand Synthesis Reaction Buffer with dUTP included in the module synthesizes the second strand of the cDNA using dUTP. Prior to the PCR enrichment step of subsequent DNA library construction, the UDG enzyme will hydrolyze the second strand cDNA to ensure that the final sequencing data was derived from the first strand cDNA, retaining the mRNA sequencing strand specificity.
